Object Description

Ocular development involves intricate network of cells derived from forebrain neuroectoderm, surface ectoderm and neural crest-derived perioptic mesenchyme (POM). Retinoic acid (RA) synthesized by retinaldehyde dehydrogenases (Raldh1, Raldh2 and Raldh3) functions in a paracrine fashion to guide morphogenetic movements in retina and POM. Pitx2, encoding homeodomain transcription factor is a direct target of RA that regulates Dkk2 (an antagonist of Wnt/b-catenin signaling) in POM to locally suppress Wnt signaling to ensure proper eye development. Raldh double null mutant (R1:R3-\-) embryos exhibit severe ocular phenotype characterized by lack of upper eyelid, disoriented optic cup, corneal dysgenesis and microphthalmia. In situ hybridization with Dkk2 riboprobe on frontal sections of E12.5 wild-type (WT) and double null (R1:R3-/-) mice eyes shows reduced expression of Dkk2 in POM of R1:R3-/- compared to WT suggesting that activated Wnt signaling could lead to these ocular defects.
